Design Notes

We digested the promoter responsive to cadmium (BBa_K174015) with EcoRI and SpeI and digested the ogr activator (BBa_I746350) with XbaI and PstI. These parts were then ligated to a Kanamycin vector that was digested with EcoRI and PstI. This created an intermediate part that was selectively grown on Kanamycin plates. This part could then be ligated to our other intermediate, BBa_K1042013, the PO Promoter and GFP Reporter. Together this would create a final construct that is theoretically a cadmium sensor with increased sensitivity.
